Cells synthesize glutathione through two ATP-dependent enzymatic steps. The first step combines glutamate and cysteine to form gamma-glutamylcysteine, catalyzed by glutamate-cysteine ligase. The second step adds glycine, producing the complete tripeptide, catalyzed by glutathione synthetase. Glutathione itself can inhibit the first enzyme, providing negative feedback when levels are high. Because cysteine is often limiting, its availability influences how quickly the pathway proceeds. These reactions occur in the cytosol, and the resulting glutathione can be distributed to other compartments.
Glutathione functions in redox balance, detoxification, and sulfur amino acid storage. It participates in reactions that help maintain ascorbate and protein thiol status. The molecule serves as a cofactor for several enzymes, including glutathione peroxidases and glutathione S-transferases. These enzymes reduce peroxides and conjugate electrophiles, respectively. Glutathione also contributes to the metabolism of xenobiotics and to the transport of cysteine between tissues. How interorgan transport and tissue-specific regulation shape whole-body pools remains an active area of study.
Synthesis occurs in two ATP-dependent steps. The enzyme glutamate-cysteine ligase joins glutamate and cysteine to form gamma-glutamylcysteine, and glutathione synthetase adds glycine. The first step is rate-limiting and is influenced by cysteine availability and feedback inhibition by GSH. Breakdown involves gamma-glutamyl transferase and subsequent peptidases, which release constituent amino acids for reuse. Because turnover differs among tissues, measurements from blood, plasma, and tissues are not directly interchangeable. Research continues to clarify how compartment-specific pools are regulated in health and disease.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. It contains an unusual gamma-glutamyl bond between glutamate and cysteine, which resists cleavage by many peptidases. The reduced form, GSH, carries a thiol group on cysteine and is the dominant intracellular form in most cells. Its structure allows it to participate in redox reactions and to serve as a sulfur donor. The oxidized form, GSSG, consists of two GSH molecules joined by a disulfide bond.
In cells, glutathione helps maintain the reducing environment of the cytosol and supports enzymes that counteract reactive oxygen species. It acts as a cofactor for glutathione peroxidases, which reduce hydrogen peroxide and lipid peroxides, and for glutathione S-transferases, which conjugate electrophiles. The ratio of GSH to GSSG is often used as an indicator of oxidative stress, although the ratio can vary by compartment and cell type. Glutathione also stores cysteine, an amino acid that can be limiting for protein synthesis and antioxidant defense.

Cells synthesize glutathione through two ATP-dependent steps: glutamate-cysteine ligase joins glutamate and cysteine, and glutathione synthetase adds glycine to form the complete tripeptide. Breakdown occurs through gamma-glutamyl transpeptidase and subsequent peptidase reactions, forming the gamma-glutamyl cycle. Within cells, glutathione also forms a disulfide-linked dimer called GSSG when two GSH molecules react. The balance between GSH and GSSG is widely used as an indicator of oxidative conditions, although the ratio can vary by compartment and tissue.
Glutathione supports several cellular processes beyond direct antioxidant action. It serves as a cofactor for glutathione peroxidase and glutathione S-transferase enzymes, which reduce peroxides and conjugate electrophiles, respectively. The molecule also acts as a reservoir of cysteine, an amino acid that can limit protein synthesis and redox signaling. In human nutrition, oral glutathione is sold as a supplement, but how much intact glutathione reaches tissues after ingestion remains an active research question. Clinical claims about supplementation are not uniformly supported by controlled trials.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. The peptide bond between glutamate and cysteine uses the gamma-carboxyl group of glutamate rather than the alpha-carboxyl group. This unusual linkage protects the molecule from many common peptidases. The cysteine side chain carries a thiol group that can undergo reversible oxidation. Because of this thiol, glutathione participates in redox reactions and helps maintain the reducing environment inside most cells in living systems.
In cells, glutathione exists mainly in a reduced form called GSH. When two GSH molecules react, they form oxidized glutathione, or GSSG, which contains a disulfide bond. The ratio of GSH to GSSG is often used as an indicator of oxidative stress. Enzymes such as glutathione peroxidase and glutathione reductase help cycle the molecule between these two states. This cycling supports antioxidant defense, detoxification of reactive molecules, and regulation of certain signaling pathways.
=== Film performances === Culp worked as an actor in many theatrical films, beginning with three in 1963: As naval officer John F. Kennedy's good friend Ensign George Ross in PT 109, as legendary gunslinger Wild Bill Hickok in The Raiders, and as the debonair fiancé of Jane Fonda in Sunday in New York. He starred in Bob & Carol & Ted & Alice in 1969, with Natalie Wood. Another memorable role came as another gunslinger, Thomas Luther Price, in Hannie Caulder (1971) opposite Raquel Welch. A year later, Hickey & Boggs reunited him with Cosby for the first time since I Spy. Culp also directed this feature film, in which Cosby and he portray over-the-hill private eyes. In 1986, he had a primary role as General Woods in the comedy Combat Academy. Culp played the U.S. President in Alan J. Pakula's 1993 murder mystery, The Pelican Brief.
== Side effects == Rasburicase administration can cause anaphylaxis (incidence unknown); methemoglobinemia may occur in susceptible individuals such as those with G6PDH deficiency due to the production of hydrogen peroxide in the urate oxidase reaction. Testing patients for G6PDH deficiency prior to starting a course of rasburicase has been recommended.
==== Road to war ==== Liu Wenhui's 1990 biography asserts that conflict between the two Lius was almost inevitable. Due to the consolidation of power in Sichuan that had occurred over the 1920s, Liu Wenhui and Liu Xiang had become the two most powerful warlords in the province. Liu Wenhui's assumption of the governorship in 1929 had turned many smaller warlords against him. By controlling the lower reaches of the Yangtze and the Qutang Gorge, Liu Xiang held the geographic advantage over his uncle, having priority in purchasing weapons from abroad (which entered Sichuan through the Yangtze), and had built a navy of three gunboats by fitting merchant ships with iron hulls. This also meant that Liu Xiang could block weapons from reaching Liu Wenhui's upstream domains, an ability that he would use to his advantage. Liu Xiang was also reportedly under the sway of his Taoist mystic advisor, Liu Congyun, who had told him that he was destined to unite the province under his rule. Yet another supposed factor contributing to conflict was the interference of Chiang Kai-shek, who did not want either warlord to become too powerful. Most sources agree that Liu Wenhui's abortive attempt to acquire military aviation assets in 1931 was the spark that led him to conflict with his nephew. Official Chinese sources state that a shipment ordered by Liu of foreign aircraft components worth 2 million yuan was seized by Liu Xiang at the port of Wanxian in Chongqing, leading to heightened tensions between the two Lius.
Merely giving a treatment can have nonspecific effects. These are controlled for by the inclusion of patients who receive only a placebo. Subjects are assigned randomly without informing them to which group they belonged. Many trials are doubled-blinded so that researchers do not know to which group a subject is assigned. Assigning a subject to a placebo group can pose an ethical problem if it violates his or her right to receive the best available treatment. The Declaration of Helsinki provides guidelines on this issue.
The molecular mass (m) is the mass of a given molecule, often expressed in units of daltons (Da). Different molecules of the same compound may have different molecular masses because they contain different isotopes of an element. The derived quantity relative molecular mass is the unitless ratio of the mass of a molecule to the atomic mass constant (which is equal to one dalton). The molecular mass and relative molecular mass are distinct from but related to the molar mass. The molar mass is defined as the mass of a given substance divided by the amount of the substance, and is expressed in grams per mole (g/mol). That makes the molar mass an average of many particles or molecules (weighted by abundance of the isotopes), and the molecular mass the mass of one specific particle or molecule. The molar mass is usually the more appropriate quantity when dealing with macroscopic (weigh-able) quantities of a substance. The definition of molecular weight is most authoritatively synonymous with relative molecular mass, which is dimensionless; however, in common practice, use of this terminology is highly variable. When the molecular weight is given with the unit Da, it is frequently as a weighted average (by abundance) similar to the molar mass but with different units. In molecular biology and biochemistry, the mass of macromolecules is referred to as their molecular weight and is expressed in kilodaltons (kDa), although the numerical value is often approximate and representative of an average.

==== Matrix-assisted laser desorption/ionization ==== MALDI is typified by the use of an ultraviolet (UV) laser to trigger ablation of analyte species that are mixed with a matrix of crystallized molecules with high optical absorption. The ions within the resulting ablated gasses are then protonated or deprotonated before acceleration into a mass spectrometer. The primary advantages of MALDI detection over ESI in microfluidic devices are that MALDI allows for much easier multiplexing, which even further increases the device's overall throughput, as well as less reliance on moving parts, and the absence of Taylor cone stability problems posed by microfluidic-scale flow rates. The speed of MALDI detection, along with the scale of microfluidic droplets, allows for improvements upon macro-scale techniques in both throughput and time-of-flight (TOF) resolution. Where typical MS detection setups often utilize separation techniques such as chromatography, MALDI setups require a sufficiently purified sample to be mixed with pre-determined organic matrices, suited for the specific sample, prior to detection. MALDI matrix composition must be tuned to produce appropriate fragmentation and ablation of analytes. One method to obtain a purified sample from droplet-based microfluidics is to end the microfluidic channel onto a MALDI plate, with aqueous droplets forming on hydrophilic regions on the plate. Solvent and carrier fluid are then allowed to evaporate, leaving behind only the dried droplets of the sample of interest, after which the MALDI matrix is applied to the dried droplets.
During October and November 2005, Wenger became embroiled in a war of words with Chelsea manager José Mourinho. Mourinho accused Wenger of having an "unprofessional obsession" with Chelsea and labelled him a "voyeur". Mourinho was quoted as saying, "He's worried about us, he's always talking about us – it's [always] Chelsea". Wenger responded by pointing out he was only answering journalists' questions about Chelsea, and described Mourinho's attitude as "disrespectful". Mourinho later apologised and clarified that he regretted his "voyeur" comment; Wenger accepted the apology. In 2014, Mourinho reopened his feud with Wenger by calling him a "specialist in failure". This was in response to Wenger's comments that Chelsea were favourites for the Premier League and managers did not want to take responsibility if they failed to win the title. Wenger said Mourinho's comments were "silly and disrespectful" and had embarrassed Chelsea. When asked if he regretted his remark, Mourinho replied: "You have to ask him if he regrets the comments he made". In October 2014, during a Premier League match between Arsenal and Chelsea, Wenger was involved in a touchline spat with Mourinho. Wenger later apologised for his behaviour. Wenger has directed his anger towards referees when decisions have not gone his team's way. In August 2000, he was charged with "alleged threatening behaviour and physical intimidation" towards fourth official Paul Taylor, after Arsenal's 1–0 defeat at Sunderland on the opening day of the 2000–01 season.

As a decentralized system, bitcoin operates without a central authority or single administrator, so that anyone can create a new bitcoin address and transact without needing any approval. This is accomplished through a specialized distributed ledger called a blockchain that records bitcoin transactions. The blockchain is implemented as an ordered list of blocks. Each block contains the hash of the previous block, made by hashing its header twice with SHA-256, chaining them in chronological order. The blockchain is maintained by a peer-to-peer network. Individual blocks, public addresses, and transactions within blocks are public information, and can be examined using a blockchain explorer. Nodes validate and broadcast transactions. Each node maintains a copy of the blockchain for ownership verification. A new block is created every 10 minutes on average, updating the blockchain across all nodes without central oversight. This process tracks bitcoin spending, ensuring each bitcoin is spent only once. Unlike a traditional ledger that tracks physical currency, bitcoins exist digitally as unspent outputs of transactions.
Finally, a rather rare and exotic side reaction is known as cine substitution. Here, after initial oxidative addition of an aryl halide, this Pd-Ar species can insert across a vinyl tin double bond. After β-hydride elimination, migratory insertion, and protodestannylation, a 1,2-disubstituted olefin can be synthesized.
Other 1,2- and 1,3-dithiols give related 1,3-dithiolanes (five-membered) and 1,3-dithianes (six-membered rings). Diols such as ethylene glycol undergo analogous reactions to give 1,3-dioxolanes. One distinguishing feature of the dithiolanes and dithianes derived from aldehydes is that the methyne group can be deprotonated and the resulting carbanion alkylated. 1,2-Ethanedithiol has been used as a scavenger in peptide cleavage synthesis. Like 1,3-propanedithiol, 1,2-ethanedithiol readily forms metal thiolate complexes. Illustrative is the synthesis of the derivative diiron ethanedithiolate hexacarbonyl upon reaction with triiron dodecacarbonyl: Fe3(CO)12 + C2H4(SH)2 → Fe2(S2C2H4)(CO)6 + H2 + Fe(CO)5 + CO Ethane-1,1-dithiol

Glutathione is built from three amino acids: glutamate, cysteine, and glycine. The linkage involves the gamma-carboxyl group of glutamate rather than the alpha-carboxyl group, which is unusual for peptides. This structure protects the bond from some common peptidases.
It is present in nearly all cells, with notable amounts in the liver, kidneys, and red blood cells. The highest intracellular concentrations are usually in the millimolar range. Levels differ by tissue, age, and physiological state.
It is not classified as an essential nutrient because cells can synthesize it from amino acids. Dietary sources exist, but their contribution to tissue pools is not fully established. The body's production depends on enzyme activity and precursor availability.
GSH is the reduced form with a free thiol group, while GSSG is the oxidized disulfide-linked dimer. Most assays distinguish the two because their balance reflects redox conditions. The names are not interchangeable.
